Does Nethereum work with C# 6 and .NET 4.6? I would like to try an integrate this into a Unity project.

there are specific libraries for Unity3d
check the release page
those are net351
to be compatible with all the versions of unity

I'm really new to all this, is the release page the same as the GitHub page? Nethereum will allow me to make my application communicate with the entherium blockchain and send/receive contracts using C# correct?

yes
for unity you need the 2.0.0 zip file (it is not in nuget)
for a sample of usage you can have a look at
and for guidance on the sample check
also you can play the game here

simple thing (i guess) is the blog post
to get started
then look at the sample
and from there get the zip file with the libraries

If I build to PC will the user require any additional software to run the application?
Or just an eth address.

you can connect to infura
so it wont need anything else
apart from their keys to sign transaction and yes an address to a contract to interact with
the game sample connects to infura and uses metamask for key management
but you can do the same in pc with just nethereum
for key management

So metamask is a browser specific plugin.
